Technology and Capabilities Questions

How does the ProTrap clean samples?

The ProTrap uses organic solvent precipitation, selectively aggregating the protein while buffer components and the sample matrix remain in solution. The solution phase is washed away by centrifugation, isolating the precipitated protein on the filtration membrane. Our protocols are based on methods published by our co-founder and R&D collaborator, Dr. Alan Doucette, Dalhousie University, and other researchers in peer-reviewed research articles.

What type of samples can be used in the ProTrap?

The ProTrap XG and ProTrap HT are compatible with clarified cell or tissue lysates as well as biofluid samples. Our team is actively working on sample processing protocols to provide users with more comprehensive parameters (e.g. more diverse buffer components) for robust precipitation. Low molecular weight proteins (<5kDa) exhibit reduced recovery. however, we are currently exploring new strategies to maximize the recovery of these smaller analyes.

How much protein can the ProTrap XG prepare?

The ProTrap XG base protocols are optimized to process 50 micrograms of protein. Our research team has successfully recovered microgram quantities of protein down to 2 micrograms and the ProTrap works well with 20-100 micrograms. If you need to process more than 50 micrograms or less than 2 micrograms of protein please contact us at support@allumiqs.com

Does the ProTrap do peptide cleanup?

Our team is currently exploring new cleanup strategies in low molecular weight (<5 kDa) proteins and peptides through precipitation. We currently recommend complementing the ProTrap workflow with a post-digestion SPE clean-up of your choosing.

Is the Serum/Plasma Sample Prep Kit limited to serum/plasma?

No, the reagents are amenable to the preparation of any clarified lysate or biofluid. The protocol provided with the kit, however, recommends sample loading and digestion buffer quantities that are optimized for serum/plasma samples.

Which MS platforms does Allumiqs employ?

We are always implementing the latest LC-MS instruments in our lab to get the most out of every sample and acquisition.  We currently run customer samples in SWATH DIA mode on our SCIEX TripleTOF 6600+ and our ZenoTOF 7600 platforms. Both are couple to microflow HPLC systems.

What other technology does Allumiqs use in customer projects?

We have recently added the Seer Proteograph™ Product Suite to our lab, expanding our depth of coverage for species-agnostic plasma proteomics.

We are currently developing a platform to encompass our proprietary data analysis tools.

Do you sell reagent kits?

For small batches, pilot projects, or CROs enabling their customers with sample prep tools we offer the Serum/Plasma Sample Prep Kit.  The kit contains ProTrap XG filtration cartridges and reagents for processing up to 10 samples.  Find out more about the Kit here.

Do you have an automated solution?

We offer the ProTrap HT 96-Well Plate for high throughput and large-scale sample processing.  Learn more about the ProTrap HT here.

What automation platforms are compatible with the ProTrap HT 96-Well Plate?

The ProTrap HT is compatible with any A200 workflow. It relies on solvent delivery and positive pressure systems to automate the entire workflow, though buffers may alternatively be exchanged via centrifugation or negative pressure.
